[摘要]目的:探讨A型肉毒毒素对人瘢痕疙瘩成纤维细胞凋亡的影响。方法:体外分离和培养人瘢痕疙瘩成纤维细胞,实验组在细胞培养过程中加入A型肉毒毒素(稀释成5U/0.1ml)进行干扰,空白对照组培养液中不加A型肉毒毒素。培养72h后利用流式细胞仪检测细胞凋亡情况。结果:3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐[3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide, MTT]法显示A型肉毒毒素作用后细胞增殖速度变慢;流式细胞仪检测显示实验组早期凋亡和晚期凋亡的细胞数量较空白对照组均明显增多,成纤维细胞凋亡率差异有统计学意义(P http://

Effects of Botulinum Toxin Type A on Apoptosis of Human Keloid Fibroblast

Abstract: Objective To investigate the effects of botulinum toxin type A (BTXA) on apoptosis of human keloid fibroblast. Methods Human keloid fibroblast was isolated and cultured in vitro, the medium containing BTXA(diluted concentration of 5U/0.1ml)was used to intervene the cell growth in experience group, BTXA was not added in blank control group. At 72 hours after cells were cultured, cells apoptosis were measured by flow cytometry( FCM ). Results Cells proliferation rate was slow with MTT method. The cell numbers of early and late apoptosis in experience group were significantly more than those of early and late apoptosis in blank control group with FCM, showing a significant difference between them(P 2.3 观察肉毒毒素对瘢痕疙瘩成纤维细胞凋亡的影响:收集第4代成纤维细胞,分成6组,实验组和空白对照?M各3组。实验组加入含A型肉毒毒素5U/0.1ml的1%胎牛血清DMEM,空白对照组培养液中不加A型肉毒毒素。72h后流式细胞仪检测,观察细胞凋亡情况:六孔板中的细胞用PBS洗涤一次,加入1ml 0.25%胰酶消化细胞,待细胞变圆且有部分细胞悬浮时加入PBS终止消化。用移液枪轻轻吹打细胞,使细胞悬浮。收集于流式管中,离心、弃上清。加入3ml 4℃预冷的PBS完全重悬细胞,1 500r/min离心5min,弃上清,沉淀用300μl的Binding Buffer重悬。加入5μl Annexin V-FITC混匀后,再加入5μl Propidium Iodide混匀。室温下避光反应5min。于1h内进行流式细胞仪检测。
2.4 统计学分析:采用SPSS13.0软件进行统计分析,两组间的细胞光密度值比较采用两样本t检验,两组间细胞凋亡率比较采用χ2检验。以P参考文献]
