2018四种食源性肽改善光老化大鼠皮肤皱纹及屏障功能的实验研究

  [摘要]目的:?u价四种食源性肽口服暴露对SD大鼠光老化皮肤皱纹及屏障功能的改善效果。方法:将112只健康成年雌性SD大鼠随机平均分为空白对照组、模型组、肽样品组,每种肽分别设定低、中、高剂量组,配制浓度为0.3、0.9、1.5g/L的含肽水溶液进行口服,每日自由饮用;空白组不施行紫外辐照,模型组及肽样品组均行UVA+UVB联合辐照,连续辐照18周后进行皮肤表观皱纹评分,检测皮肤表皮水分含量和油脂含量,评价4种食源性肽对光老化大鼠皮肤皱纹和屏障功能的改善效应。结果:与正常对照组相比,模型组大鼠皮肤皱纹感官质量显著降低,肽口服干预后可不同程度改善皮肤皱纹状况,且总体呈剂量-效应关系,不同活性肽之间改善效果差异较大,以样品2高剂量组效果最好;从屏障功能来看,模型组大鼠皮肤水分和油脂含量显著低于空白组(P http://

  Abstract: Objective To evaluate the improvement of four kinds of food-derived peptides with oral supplementation on wrinkling and barrier function status of SD rat photoaging. Methods 112 of healthy adult female SD rats were randomly divided into normal control group, ultraviolet exposed group and peptides administration groups. Except the normal group, all animals in model and peptides administration groups were exposed to radiation of UVA in companying with UVB. The low, middle and high-dose bioactive peptides administration were obtained by preparing the water solution of peptides with concentration of 0.3, 0.9 and 1.5g/L respectively. And then the obtained peptides solution was orally administrated to the SD rats in peptides treating groups as the sole drinking source. The model and peptides groups were exposed to UV irradiation thrice weekly for 18 weeks until the photoaging model was established and then the improvement of four kinds of food-derived peptides on wrinkling and barrier function was evaluated by sensory scoring and operatically detection. Results In comparison with the rats in normal control group, the animals in model groups had remarkable wrinkling formation and this degradation in wrinkling status was improved by oral supplementation of peptides and the effectiveness was in a dose-effect manner and dependent on the kind and concentration of, of which the sample of peptides 2 exhibited most distinguished wrinkling elimination. Furtherly, judging from the water and oil barrier function of skin, the moisture and lipid concentration of skin in model group were significantly lower than those in normal group and the barrier function was desirable improved by oral supplementation of peptides (P    Key words: peptides; oral supplementation; SD rats; photoaging; barrier function
  皮肤老化与抗衰老是当今皮肤科学的研究热点,紫外线辐射引起的皮肤光老化是皮肤外源性衰老的主要原因[1]。光老化导致皮肤过早出现粗糙、皱纹、色素异常、脆性增加、毛细血管扩张及皮革样外观等症状,严重影响皮肤外观,导致皮肤屏障功能受损[2-4]。随着审美标准的不断提高,人们对皮肤光老化的防治越来越重视,鉴于皮肤组织的天然“砖墙结构”的致密屏障,只有分子量小于3 000Da以下的分子才能透皮吸收,而且功效成分大多化学结构稳定性较差,长时间暴露于紫外线等环境中极易分解,失去本身应有的抗衰功能甚至产生有毒物质[5]。因此,通过饮食等内在途径改善皮肤屏障功能,减少皮肤皱纹量,从而改善光老化皮肤表观形态已是新一代抗衰功能食品开发的必然趋势。本研究对四种食源性肽进行口服实验,根据光老化皮肤皱纹改善及屏障功能恢复情况,筛选最佳食源性肽,为抗衰产品开发奠定实验基础。
  1 材料和方法
  1.1 材料和试剂:健康雌性SPF级SD大鼠168只购自广州中医药大学实验动物中心提供,周龄10周,体质量180~200g,实验动物生产许可证号:SCXK(粤)2011-0029,质量合格证号:No.44008500003344。
  食源性肽样品1、2、3由无限极(中国)有限公司技术中心提供(样品1为狭鳕皮肤胶原蛋白肽,购自广州维晟生物科技有限公司;样品2为牛源弹性蛋白肽,由无限极(中国)有限公司技术中心自制;样品3为鲣鱼动脉球弹性蛋白肽,购自 Hayashikane Sangyo Co,.Ltd),核桃肽由华南理工大学赵谋明教授惠赠。新鲜基础鼠粮及清洁刨花垫料购自广州市花都区实验动物养殖场。
  1.2 主要设备:紫外光辐照箱为本实验室自行设计安装[6],已获专利授权(专利号:ZL201520152906.5)。紫外线灯管:UVA Philips 340-40WT12-G13,波长范围320~400nm,波峰340nm;UVB Philips 313-40WT12-G13,波长范围300~320nm,波峰313nm。UV-A型、UVB297型紫外辐照计购自北京师范大学光电仪器厂。大鼠实验笼购自苏州市苏杭科技器材有限公司。FC1502菲斯?凯尔皮肤分析仪来自深圳凯尔电子厂。
  1.3 方法
  1.3.1 动物分组与模型建立:SD大鼠饲养在专用鼠笼中,环境设施符合试验动物SPF级等级要求,室温20℃~25℃,湿度40%~70%,自然昼夜交替。动物实验遵守实验动物伦理学要求,大鼠适应性喂养1周后,最终选取112只健康SD大鼠随机分为14组,正常对照组、模型组和肽样品组(4种肽样品分为设置低、中、高剂量组),每组8只,各组大鼠分2笼饲养,每笼4只。正常对照组与模型组以清洁自来水为唯一饮用水,肽样品低、中、高剂量组分别在清洁自来水中分别加入终浓度0.3、0.9、1.5g/L的样品1、2、3及核桃肽,饲料均为基础饲料,自由摄食。
  正常对照组不进行辐照,其余各组进行等量紫外辐照。参照文献[6-9],用记号笔画出背部脊柱两侧约5.0cm×5.0cm区域并用电推剪剃除毛发,每次辐照前剃毛并?M行Na2S化学脱毛,将脱毛大鼠放入自制固定器中暴露背部剃毛皮肤,连同固定器和大鼠置于紫外线光源(UVA+UVB)正下方40cm处进行紫外辐照,紫外辐照计测得该处UVA强度为177.9mW/cm2,UVB强度为59.6mW/cm2。辐照频率为隔天辐照1次,时间为第1周10min/d,第2周20min/d,第3周30min/d,之后维持在30min/d,第18周皮肤外观呈典型老化状态表示造模结束,累计照射剂量为UVA 150.88J/cm2,UVA 78.67J/cm2,紫外辐照结束后对各组大鼠进行观察拍照。
  1.3.2 皮肤表观皱纹测定评分:参照文献资料[10],根据皮肤横向皱纹的情况对光老化程度和改善效果进行评分,评分依据见表1。
  1.3.3 皮肤屏障功能测定:皮肤屏障功能评价采用皮肤表皮水分含量和油脂含量进行量化,FC1502菲斯?凯尔皮肤分析仪进行测定,具体按照仪器说明书进行操作,采用点触式测量,测3次取平均值。
  1.4 统计学分析:使用SPSS16.0软件对数据进行统计分析,实验数据以均数±标准差(x?±s)表示。组间比较采用单因素方差分析,两两比较方差齐时采用LSD检验法,方差不齐时采用Dunnett’s T3 进行比较,P    2.3 各组SD大鼠皮肤屏障功能测定结果:由图3A可以看出,与正常对照组相比,模型组SD大鼠皮肤在光老化造模过程中表层水分含量显著降低(P参考文献]
